Xcellerex offers two options for cell line development:

Clone Screening

High-throughput (HT) clone screening is a platform which allows a variety of cell types to be rapidly screened and cloned after transfection to select high producers. Millions of cells can be screened, reducing cell line development time and increasing the odds of isolating the highest producers.

High-throughput clone screening was created because transfection is a random event. The gene of interest integrates randomly into the genome of the host cell. The vast majority of these recombination events occur in loci which are "silent" or disrupt the metabolism of the cell. Researchers typically screen hundreds to thousands of clones in order to isolate a few clones with an acceptable level of productivity. Once isolated, these clones are typically carried through multiple rounds of limited dilution cloning to assure a truly clonal cell line.

For mammalian cells, each round of cloning takes several weeks. Xcellerex has developed a high-throughput clone screening system that can rapidly screen millions of cells and reduce cloning times by months. When combined with the high-throughput protein titer assay, these techniques have the potential to reduce production cell line generation time by months and screen over 1000 times more clones for high producers than manual methods, resulting in the best chance to find the highest producers.
